Fluoride is widely used as a component of toothpaste for the prevention of dental caries. In this study, the fluoride >k effect on the proliferation of human gingival fibroblast in culture system was investigated. NaF at low concentrations (50 M-500 pM) stimulated cell proliferation. But, at higher concentrations(over 500 pM) gradually reduced the cell growths AJ.CL at low concentrations(5-20 pM) potentiated the cell growth stimulated by NaF. NaF induced the formation of inositol 1, 4, 5 trisphosphate in concentration-dependent manner. A1CL potentiated the IP3 formation induced t by NO.
These data suggested that the human gingival cell proliferation stimulated by fluoroaluminate, the combined from of Al-"- and F-, may be due to the activation of phosphoinositide-specific phospholipase C through guanine nucleotide-binding protein.
